Hi all,

I’m looking for good datasets, analyses, or papers on surgicogenomics in PD—especially how variants like _GBA1_ (e.g., p.L444P) may influence DBS outcomes (motor response, cognition, complications) or target selection.

I’m working on a case that raised this question, and I’d like to base our approach on solid data as we start integrating genetics more formally into DBS decisions.

Any pointers or work in this area would be really helpful.

Thanks!

Hi @psaffie, good question and sounds like a really intriguing line of inquiry. Tagging community members who have experience / interest in DBS to see if they have any input!

Hello!

I know that PPMI has a genetic cohort, and in general, some patients had DBS surgery at some point of the study. There are columns that tell you whether they had surgery and the date, so you can get some post-surgical prospective data.

I am not sure how many patients with GBA had surgery, but according [to this paper](https://pubmed.ncbi.nlm.nih.gov/36790548/), there are at least 44 GBA patients by 2021. They recently added more data in late 2025, so it might be more now.

Good luck!

it looks like an Italian team has done some work on this if you’d like to reach out to them?

> The impact of DBS on clinical outcomes in GBA-PD patients was unexplored until few years ago. Recent studies showed that DBS improves long-term motor function without affecting cognition in GBA-PD.
