I’m looking for good datasets, analyses, or papers on surgicogenomics in PD—especially how variants like GBA1 (e.g., p.L444P) may influence DBS outcomes (motor response, cognition, complications) or target selection.
I’m working on a case that raised this question, and I’d like to base our approach on solid data as we start integrating genetics more formally into DBS decisions.
Any pointers or work in this area would be really helpful.
Hi @psaffie, good question and sounds like a really intriguing line of inquiry. Tagging community members who have experience / interest in DBS to see if they have any input!
I know that PPMI has a genetic cohort, and in general, some patients had DBS surgery at some point of the study. There are columns that tell you whether they had surgery and the date, so you can get some post-surgical prospective data.
I am not sure how many patients with GBA had surgery, but according to this paper, there are at least 44 GBA patients by 2021. They recently added more data in late 2025, so it might be more now.
